The Fluorescence Microscope How To Prepare Specimen For The Fluorescence Microscope Fluorescence microscopy is one of the most common approaches in studying the location and movement of molecules and subcellular components in the cell. Why Prepare The Specimen? Most cells are colorless by nature; therefore, fluorescent markers are introduced in them to make them easier to study. 3 Here are 3 techniques that are used to #1 prepare specimen for the Fluorescence Microscope. Fluorescent Dyes Fluorescent dyes are incorporated and concentrated in specific subcellular compartments, as they are directly taken up by the cells. The living cells are then mounted on a microscope slide and examined in a fluorescence microscope. #2 Tagging of Proteins Protein cells can be modified to create their own fluorescent molecules by tagging them with a fluorescing marker. When a specific protein is modified in this way, the location of that protein can be studied. It is also possible to watch the movements of the proteins and its interactions with other cellular components inside the cell. #3 Immunofluorescence Immunofluorescence involves introducing antibodies tagged with a fluorescing marker. Antibodies are molecules that recognize and bind selectively to specific target molecules in the cell.
